As for the logic of who's big and who's petite... well, just look at the humanized versions of most of these characters and you start to see the logic. Pinkie's gluttony and rubbery hijinks makes it easy to imagine her as 'big'. Rainbow is a racer, and most female olympic runners in real life are pretty small in the chest area, and so forth.
But yeah, as you said with the joke in Futurama, I was wondering if there was any difference.
